Clinical research reveals: NMN’s effect on metabolism

How can NMN contribute to better metabolic health? A research team from Washington University School of Medicine in St. Louis investigated this topic in a clinical study published in the journal Science. The study focuses on NMN’s potential to improve metabolism in postmenopausal women aged 55 to 75 with prediabetes.

Background of the study

Aging and obesity often cause a decline in NAD+ levels, which play a critical role in energy production and metabolism. Previous animal studies have shown that NMN can boost NAD+ levels, helping to address metabolic issues like insulin resistance and impaired glucose tolerance. The study we’re examining is the first to explore how NMN affects metabolism in humans.

How the researchers conducted the study

The researchers conducted a 10-week randomized, double-blind, placebo-controlled trial with 25 postmenopausal women who had prediabetes and a BMI between 25.3 and 39.1. They split the participants into two groups:

  • NMN group: Took 250 mg of NMN daily for 10 weeks.
  • Placebo group: Took a placebo daily for 10 weeks.

What was measured

At the beginning and end of the study, researchers measured:

• Muscle cell changes: Muscle samples were analysed to identify any differences in the cellular function.

• Body fat and muscle mass: To determine if there were changes in body composition.

• Insulin sensitivity: Assessed using a hyperinsulinemic-euglycemic clamp test to see how well the body responded to insulin.

The results of the study

• NAD+ and NMN Metabolites: The group of women who took NMN showed an increase in NAD+ levels in blood cells and metabolites linked to NAD+ turnover in muscles. The placebo group showed no changes.

Insulin Sensitivity: The NMN group experienced a significant improvement in muscle insulin sensitivity, with an increase of 25% compared to baseline. No changes were observed in insulin sensitivity in the liver or fat tissue. The placebo group showed no changes.

Gene Expression: NMN significantly increased the number of active genes during insulin infusion (308 genes in the NMN group compared to only 5 in the placebo group). The researchers found that these genes were mainly linked to tissue repair and regeneration.

No Changes in Body Weight or Composition: There were no significant changes in body weight, fat mass, or muscle mass in either group after 10 weeks of treatment.

Safety: The researchers reported no side effects, and all standard blood tests remained normal.

Implications of the Results

This is the first clinical study to document that NMN can improve insulin sensitivity in humans. The findings emphasize NMN’s potential as a targeted treatment for age-related metabolic disorders and prediabetes, particularly in postmenopausal women, who are often at higher risk for these conditions.

The study demonstrates that NMN can play an important role in enhancing muscle function by improving blood sugar regulation and promoting tissue repair. This may contribute to the prevention of type 2 diabetes and reduce the long-term complications associated with insulin resistance.

The positive results, combined with a strong safety profile and absence of side effects, make NMN a promising candidate for further research. Future studies could investigate whether NMN also benefits other tissues, such as the liver and fat, and explore whether longer treatment durations and higher doses may amplify its effects.

This paves the way for new approaches to managing metabolic diseases and offers hope that NMN could support healthy aging and improve quality of life.

Other results on the muscle function with NMN

A study conducted by the University of Tokyo in 2022 investigated the effects of 250 mg of NMN daily for 12 weeks on men aged 65 and older. The results showed that NMN significantly enhanced muscle strength, improving grip strength, walking speed, and the ability to stand up from a chair within 30 seconds. Researchers found the supplement to be well-tolerated and safe.

More positive results on insulin sensitivity with NMN

A recent review of multiple studies confirms NMN’s benefits for muscle function, liver health, and insulin sensitivity in aging adults. NMN supplementation improved walking speed by up to 20%, boosted liver function by reducing key enzymes by 15%. The study also showed that NMN improved insulin sensitivity by 8-10%, potentially lowering the risk of developing diabetes. The researchers found the supplement to be well-tolerated, with no serious side effects reported. These findings highlight NMN’s role in supporting muscle strength, metabolic health, and overall vitality.
